目的研究凹顶藻萜类化合物(LET)与VE合用对酒精诱导肝损伤大鼠的肝功能、血脂、肿瘤坏死因子(TNF-α)水平的影响,并探讨其可能的作用机制。方法70只雄性Wistar大鼠随机分为7组。酒精模型组每天酒精12ml/kgbw灌胃;阴性对照组给予等体积蒸馏水;VE干预组每天VE30mg/kgbw灌胃;LET低、中、高剂量合用干预组每天分别LET25mg/kgbw和VE30mg/kgbw、LET50mg/kgbw和VE30mg/kgbw、LET100mg/kgbw和VE30mg/kgbw灌胃;甘草酸二胺胶囊干预组给予甘草酸二胺胶囊200mg/kgbw灌胃。除阴性对照组外,其余各组酒精剂量同模型组。实验进行7周。分别测定肝功、血脂、TNF-α水平。结果与阴性对照组比较,酒精模型组血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、甘油三酯(TG)、胆固醇(CHO)、TNF-α水平差异有统计学意义(P<0.05);与酒精模型组比较,LET中、高剂量合用组及甘草酸二胺胶囊干预组ALT、TG水平差异有统计学意义(P<0.05);中、高剂量合用干预组AST水平及高剂量合用干预组CHO水平差异有统计学意义(P<0.05)。高剂量合用干预组及甘草酸二胺胶囊组TNF-α水平差异有统计学意义(P<0.05)。结论LET与VE合用能调整酒精性肝损伤大鼠体内脂质代谢,改善肝功能,降低TNF-α的水平。
Objective To investigate the effects of combination of terpenoid (LET) and VE on hepatic function, blood lipids and tumor necrosis factor (TNF-α) in alcohol-induced liver injury and to explore its possible mechanism. Methods Seventy male Wistar rats were randomly divided into seven groups. Alcohol group was given intragastric administration of alcohol 12ml / kg bw daily; negative control group was given equal volume of distilled water; VE intervention group daily VE30mg / kgbw intragastric administration; LET low, medium and high dose combined intervention group LET25mg / kgbw and VE30mg / / kg bw and VE30mg / kgbw, LET100mg / kgbw and VE30mg / kgbw intragastric administration; glycyrrhizic acid diamine capsule intervention group was given glycyrrhizic acid diamine capsule 200mg / kgbw intragastric administration. In addition to the negative control group, the remaining groups of alcohol dose with the model group. Experiment for 7 weeks. Liver function, blood lipids and TNF-α were measured respectively. Results Compared with the negative control group, serum ALT, AST, triglyceride (TG), cholesterol (CHO) and TNF-α in alcohol model group had significant difference (P <0.05). Compared with the alcohol model group, the levels of ALT and TG in middle and high dose LET group and glycyrrhizic acid diamine capsule group were significantly different (P <0.05) There was significant difference in CHO level between high and low dose intervention groups (P <0.05). There was significant difference in TNF-αlevel between high-dose combined intervention group and glycyrrhizic acid diamine capsule group (P <0.05). Conclusion LET combined with VE can regulate lipid metabolism, improve liver function and reduce the level of TNF-α in alcoholic liver injury rats.
