目的研究加味生脉饮含药血清对非小细胞肺癌细胞迁移、黏附的影响,并探讨其分子机制。方法以生药量18 g/kg灌胃家兔,制备加味生脉饮含药血清,同容积生理盐水灌胃制备空白血清。体外培养细胞,收集对数期生长的细胞,设10%空白血清组,2.5%、5%、10%含药血清组4个组。Transwell和黏附实验分别检测加味生脉饮含药血清对非小细胞肺癌细胞迁移及黏附的影响;RT-PCR测定经含药血清处理后细胞中N钙连素(N-cad)及波形蛋白(vimentin)基因表达的变化。结果随着含药血清浓度的增高,迁移到膜下表面的细胞数减少,5%和10%含药血清组迁移率仅为25.64%和13.31%(P<0.01)。随着含药血清浓度增高和作用时间延长,异质黏附率降低,且黏附率和血清浓度及作用时间呈负相关;同时N-cad、vimentin基因的表达量下降,呈浓度与时间相关性。结论加味生脉饮含药血清对非小细胞肺癌细胞有抑制迁移和降低异质黏附作用,其分子机制与影响上皮间质转化有关。
Objective To study the effect of Jiawei Shengmaiyin containing serum on the migration and adhesion of non-small cell lung cancer cells and to explore its molecular mechanism. Methods Rabbits were fed with crude drug dose of 18 g / kg, and the serum containing Jiamai Shengmai Yin was prepared. The same volume of normal saline was used to prepare blank serum. Cells were cultured in vitro and cells in logarithmic phase were collected. 10% blank serum group, 2.5%, 5% and 10% drug-containing serum group were divided into 4 groups. Transwell and adhesion experiments were used to detect the effects of Jiawei Shengmaiyin containing serum on the migration and adhesion of non-small cell lung cancer cells. RT-PCR was used to detect the expression of N-cad and vimentin vimentin) gene expression changes. Results With the increase of serum concentration, the number of cells migrated to the lower surface of the membrane decreased. The mobility of 5% and 10% drug-containing serum group was only 25.64% and 13.31% (P <0.01). With the increase of drug-containing serum concentration and the prolongation of action time, the rate of heterogeneous adhesion decreased, and the adhesion rate was negatively correlated with the serum concentration and time. At the same time, the expression of N-cad and vimentin was decreased. Conclusion Jiaweishengmai medicated serum can inhibit the migration and reduce the heterogeneous adhesion of non-small cell lung cancer cells. The molecular mechanism is related to the influence of epithelial-mesenchymal transition.
