目的:研究人单核/巨噬细胞系THP-1与人原代脂肪细胞共培养体系中白细胞介素(IL-6)分泌水平的变化,以探讨炎症和肥胖的相关性。方法:建立THP-1与人原代脂肪细胞共培养体系,并用JNK抑制剂干预共培养体系24h后,用酶联免疫吸附法(ELISA)分别测定单核细胞组、脂肪细胞组、共培养组及共培养加JNK抑制剂组上清中IL-6的水平。结果:共培养组较单核细胞组、脂肪细胞组IL-6水平升高(P<0.05),共培养加入JNK抑制剂后,IL-6水平有所下降(P=0.114)。结论:人单核/巨噬细胞系THP-1与人原代脂肪细胞共培养促进炎症因子IL-6的释放,选择性阻断炎症介导的信号传导途径,IL-6的释放减少,对症使用抗炎介质及JNK抑制剂有望成为肥胖治疗的新靶点。
OBJECTIVE: To study the changes of interleukin (IL-6) secretion in human monocyte / macrophage THP-1 co-culture system with human primary adipocytes to explore the relationship between inflammation and obesity. Methods: The co-culture system of THP-1 with human primary adipocytes was established and the co-culture system of THP-1 and human primary adipocytes was established. After intervention with JNK inhibitor for 24 hours, the number of monocytes, adipocytes and co-culture groups were measured by enzyme linked immunosorbent assay (ELISA) And co-culture plus JNK inhibitor group supernatant IL-6 levels. Results: IL-6 level in co-culture group was higher than that in monocyte group and adipocyte group (P <0.05). IL-6 level decreased after co-cultured with JNK inhibitor (P = 0.114). CONCLUSION: THP-1, a human monocyte / macrophage cell line co-cultured with human primary adipocytes, promotes the release of the inflammatory cytokine IL-6 and selectively blocks inflammatory-mediated signaling pathways, decreases in the release of IL-6 and symptomatic The use of anti-inflammatory mediators and JNK inhibitors is expected to become a new target for obesity treatment.
