目的:通过研究补骨脂酚对人真皮正常成纤维细胞(ESF-1)增殖活性以及抗老化相关蛋白mRNA表达水平的影响,探讨补骨脂酚延缓皮肤衰老的机制。方法:将体外培养的正常ESF-1细胞分为空白对照组、雌二醇(10-3μmol/L)阳性对照组及补骨脂酚高、中、低剂量(100、10、1μmol/L)组,采用MTT法检测细胞活性,逆转录聚合酶链式反应(RT-PCR)法检测药物作用后细胞中Ⅰ型胶原蛋白(ColⅠ)、Ⅲ型胶原蛋白(ColⅢ)、基质金属蛋白酶抑制剂1(TIMP-1)、基质金属蛋白酶抑制剂2(TIMP-2)、基质金属蛋白酶1(MMP-1)mRNA的表达水平。结果:雌二醇和中剂量补骨脂酚能促进ESF-1细胞的增殖,显著上调ColⅠ、ColⅢ、TIMP-1、TIMP-2 mRNA的表达,下调MMP-1 mRNA的表达。结论:补骨脂酚能促进ESF-1细胞的增殖,促进胶原蛋白和基质金属蛋白酶抑制剂mRNA的表达,并能抑制基质金属蛋白酶mRNA的表达,从而发挥抗皮肤衰老的作用。
OBJECTIVE: To investigate the effect of bakuulool on the proliferation of human dermal fibroblasts (ESF-1) and the expression of anti-aging-related protein mRNA in order to explore the mechanism of bakuyangol’s delaying skin aging. Methods: Normal ESF-1 cells cultured in vitro were divided into blank control group, estradiol (10-3μmol / L) positive control group and bakuyiphenol high, medium and low dose (100,10,1μmol / L) The cell viability was measured by MTT assay. The expressions of ColⅠ, Col Ⅲ, MMP-1 and TIMP-1 in the cells were detected by reverse transcriptase-polymerase chain reaction (RT-PCR) (TIMP-1), tissue inhibitor of metalloproteinase 2 (TIMP-2) and matrix metalloproteinase-1 (MMP-1) Results: Estradiol and middle dose of bakuyiphenol could promote the proliferation of ESF-1 cells and up-regulate the expressions of ColⅠ, ColⅢ, TIMP-1 and TIMP-2 mRNA and down-regulate the expression of MMP-1 mRNA. CONCLUSION: Bakuchiol can promote the proliferation of ESF-1 cells, promote the expression of collagen and matrix metalloproteinase inhibitor mRNA, and inhibit the expression of matrix metalloproteinase mRNA, thereby exerting anti-skin aging effect.
