目的 获取具有抗原性的旋毛虫新生幼虫基因。 方法 以旋毛虫感染猪血清为抗体探针 ,对旋毛虫新生幼虫cDNA文库进行免疫筛选 ,将获得的阳性克隆进行测序及分析。 结果 从 4 5× 10 5旋毛虫新生幼虫cDNA文库重组子中共获得 15 8个阳性克隆。序列分析结果表明 ,其中有 3 6个新的cDNA序列 ,已报道的序列有 5个 ,有 12个序列无开放阅读框架 (ORF) ,与旋毛虫线粒体基因组DNA同源。 结论 获得具有抗原性的旋毛虫抗原编码cDNA分子
Objective To obtain the antigenic Trichinella spiralis larvae gene. Methods Trichinella spiralis neonatal larval cDNA library was screened by immunization with swine serum of Trichinella spiralis as an antibody probe. The positive clones obtained were sequenced and analyzed. Results A total of 15 8 positive clones were obtained from 45 × 105 Trichinella spiralis newborn cDNA library recombinants. Sequence analysis showed that there were 36 new cDNA sequences, 5 reported sequences and 12 sequences without ORF, which were homologous to the Trichinella mitochondrial DNA.
