目的探讨hsa-miR-20在胶质瘤中的表达情况及对胶质瘤细胞增殖的影响。方法应用实时荧光定量逆转录聚合酶链反应方法检测正常脑组织和不同级别脑胶质瘤组织中miR-20a的表达情况;脂质体转染miR-20a模拟物后,通过MTT比色法及流式细胞术观察胶质瘤U251细胞增殖的变化。结果 (1)与正常脑组织相比,胶质瘤组织miR-20a的表达水平显著增高(P=0.035);(2)胶质瘤组织恶性程度越高,miR-20a表达越高(P<0.001);(3)u251细胞转染miR-20a模拟物后,增殖能力明显加快,S期细胞比例明显增多(P<0.001)。结论 miR-20a能促进胶质瘤细胞的增殖,其高表达在脑胶质瘤的发生、发展中起重要作用。
Objective To investigate the expression of hsa-miR-20 in glioma and its effect on the proliferation of glioma cells. Methods Real-time fluorescent quantitative reverse transcription-polymerase chain reaction was used to detect the expression of miR-20a in normal brain tissue and glioma tissues of different grades. After transfection with miR-20a mimics, Flow cytometry was used to observe the proliferation of glioma U251 cells. Results (1) Compared with normal brain tissue, the expression of miR-20a in glioma tissues was significantly increased (P = 0.035). (2) The higher malignant degree of glioma tissue, the higher the expression of miR- 0.001). (3) After transfected with miR-20a mimics, u251 cells proliferated significantly and the proportion of S phase cells increased significantly (P <0.001). Conclusion miR-20a can promote the proliferation of glioma cells, and its high expression plays an important role in the occurrence and development of glioma.
