目的 研究兔颈动脉平滑肌细胞核DNA含量随缺血时间而变化的关系,判断断指再植时限。方法7只新西兰大白兔过量麻醉法处死,常温(25℃)和冷藏(0~4℃)保存条件下,在0h、4h、8h、12h、16h和24h时取材,Feulgen染色后,图像细胞光度仪检测颈动脉平滑肌细胞核DNA含量,统计学处理数据。结果平滑肌细胞核DNA含量在常温下缺血8小时后,随缺血时间延长而较有规律的下降,缺血12h以内下降不明显,12h后下降比较明显。冷藏组(0~4℃)平滑肌细胞核DNA含量无明显变化。结论 断指再植时限常温下以12h内为宜。冷藏保存的肢(指)体24h内都可再植。
Objective To study the relationship between nuclear DNA content in rabbit carotid artery smooth muscle cells and ischemic time and to determine the time limit for replantation of severed fingers. Methods Seven New Zealand White rabbits were sacrificed by overdose anesthesia and were harvested at 0h, 4h, 8h, 12h, 16h and 24h after storage at room temperature (25 ℃) and cold storage (0-4 ℃). After staining with Feulgen, Instrument detection of carotid smooth muscle cell nuclear DNA content, statistical data. Results The content of nuclear DNA in smooth muscle cells decreased regularly at 8 hours after ischemia at normal temperature, decreased more regularly within 12 hours after ischemia, and decreased significantly after 12 hours. Cold storage group (0 ~ 4 ℃) smooth muscle cell nuclear DNA content no significant change. Conclusions The replantation time of severed finger is suitable within 12h at room temperature. Frozen limbs (fingers) can be replanted within 24h.
