目的:观察局灶性脑缺血损伤后大鼠脑组织中Semaphorin 3A(Sema3A)蛋白表达水平,以及通络救脑注射液对该因子表达的影响。方法:以线拴法制作大鼠大脑中动脉缺血模型(MCAO),设正常组、模型组及通络救脑注射液组,分别在造模后12h、24h、3d、7d,共4个时间点采取动物脑组织,测定脑梗死体积及脑组织中GAP43的表达水平,及Sema 3A的表达部位和蛋白表达水平。结果:通络救脑注射液能够减少脑缺血动物脑梗死体积,与模型组相比均有显著差异。结论:大鼠脑缺血损伤后Sema3A表达增加与神经元的坏死有相关性,通络救脑注射液能够降低其表达水平,对缺血损伤的脑组织发挥保护作用。
OBJECTIVE: To observe the expression of Semaphorin 3A (Sema3A) protein in rat brain after focal cerebral ischemic injury and the effect of Tongluo Jiunao injection on its expression. METHODS: Rat middle cerebral artery ischemia model (MCAO) was made by the method of sutures and was divided into normal group, model group and Tongluo Jiunao injection group. There were 4 at 12h, 24h, 3d and 7d after modeling. Animal brain tissue was taken at time point to determine the volume of cerebral infarction and the expression level of GAP43 in brain tissue, and the expression site and protein expression level of Sema 3A. RESULTS: Tongluo Jiunao Injection could reduce the cerebral infarction volume in cerebral ischemia animals, and there were significant differences compared with the model group. Conclusion: The increased expression of Sema3A is associated with the necrosis of neurons after cerebral ischemic injury in rats. Tongluo Jiunao Injection can reduce its expression level and play a protective role in ischemic brain tissue damage.
