目的 :研究N 乙酰氨基葡萄糖对白血病细胞K5 6 2的诱导分化效果 .方法 :利用hexamethylenebisacetamide(HMBA)和N 乙酰氨基葡萄糖 ,分别用不同浓度对白血病K5 6 2细胞进行诱导分化实验 ,通过联苯胺、萘酚AS D氯醋酸酯酶及吉姆萨 瑞氏染色、电镜观察、流式细胞仪证实其分化方向 .结果 :化学诱导剂N 乙酰氨基葡萄糖对K5 6 2细胞在0 .5mmol·L-1浓度作用 2d后有良好的诱导分化效果 ,诱导K5 6 2向巨噬细胞分化 .与HMBA相比 ,N 乙酰氨基葡萄糖在作用浓度和作用时间上都有明显优势 .结论 :N 乙酰氨基葡萄糖可能诱导K5 6 2细胞向巨噬细胞方向分化
OBJECTIVE: To study the effect of N-acetylglucosamine on the differentiation of leukemia K562 cells.Methods: Hexamethylenebisacetamide (HMBA) and N-acetylglucosamine were used to induce differentiation of leukemia K562 cells, respectively, by benzidine, Naphthol AS D chloroacetate esterase and Giemsaray staining, electron microscopy, flow cytometry to confirm the direction of differentiation.Results: The chemical inducer N-acetylglucosamine K5 6 2 cells in 0.5mmol·L-1 concentration After 2 days, the effect of inducing differentiation of K562 to macrophages was good.Compared with HMBA, N-acetylglucosamine had obvious advantages in concentration and duration of action.Conclusion: N-acetylglucosamine may induce K5 6 2 cells differentiate toward macrophages
