目的观察短期和长期给予茶多酚(TP)对+GZ作用下小鼠脑过氧化反应的抑制效应和TP的一般毒理学作用。方法①短期给予TP:小鼠ip给予TP,100mgkg-1d-1,3d,对照组给予0.9%NaCl,在+1GZ、+3GZ、+9GZ、+18GZ作用15min后测定脑中MDA的含量;②长期给予TP:小鼠灌胃给予TP,300mgkg-1d-1,6dwk-1,4wk,对照组给等量蒸馏水,在+1GZ、+18GZ作用15min后观察鼠脑MDA的变化;③TP的一般毒理学作用:在长期灌胃给予TP后,观察体重、脏重指数、血脂、肝肾功能和重要脏器组织形态变化。结果随暴露的+GZ值升高,小鼠脑MDA生成明显增加,但短期或长期给予TP均明显抑制此过氧化反应。TP无明显毒性作用,并可显著降低小鼠体重和血清甘油三酯水平。结论天然抗氧化剂TP对+GZ作用下小鼠脑脂质过氧化损伤具有明显保护作用,并且适于长期服药。
Objective To observe the inhibitory effect of short-term and long-term administration of tea polyphenols (TP) on brain peroxidation induced by + GZ in mice and the general toxicological effects of TP. Methods ① Short-term administration of TP: 100mgkg-1d-1 and 3-d were given ip in mice and 0.9% NaCl in control group. MDA contents in the brain were measured after + 1GZ, + 3GZ, + 9GZ and + TP: The mice were given intragastric administration of TP, 300mgkg-1d-1, 6dwk-1 and 4wk, and the control group was given the same amount of distilled water. The changes of MDA in the brain were observed after + 1GZ and + 18GZ for 15min. Long-term gavage given TP, observed body weight, body mass index, blood lipids, liver and kidney function and changes in vital organs. Results With the increase of + GZ exposure, the MDA level in the brain of mice increased significantly, but the short-term or long-term administration of TP significantly inhibited this peroxidation. TP has no obvious toxic effect, and can significantly reduce body weight and serum triglyceride levels in mice. Conclusion The natural antioxidant TP has obvious protective effect on lipid peroxidation damage of mouse brain under the action of + GZ, and is suitable for long-term medication.
