目的探讨止血带预处理后诱发肢体缺血再灌注损伤大鼠血浆内皮素(ET)的变化及缺血预处理对其的影响。方法将60只体质量150~200 g健康SD大鼠(雌雄不限)编号后采用随机数字表法将其分为对照组和实验组,各30只。对照组未经过缺血预处理,于直接缺血2、3、4 h及恢复血流后第1、3、7、14天取血测定血浆ET水平;实验组在大腿根部应用气囊止血造成肢体缺血后,止血带放松10 min形成缺血预处理过程,反复3次,于第2天分别缺血2、3、4 h后及恢复血流后第1、3、7、14天取血测定血浆ET水平。结果两组第1、3、7天同一处理时间血浆ET水平比较,P<0.05或<0.01;第14天两组缺血3 h血浆ET水平比较,P<0.01。实验组和对照组血浆ET水平在第1、3、7、14天内呈先递增后递减的趋势,且在第3天或者第7天时达峰值(P均<0.05)。实验组内第1、3、5、7天不同处理时间血浆ET水平比较,P<0.05或<0.01;对照组内第1、7天不同处理时间比较,P<0.05或<0.01。实验组缺血2、3、4 h后不同观察时间血浆ET水平比较,P均<0.01;对照组缺血2、3、4 h后不同观察时间比较,P均<0.01。结论在大鼠肢体再灌注损伤时,血浆ET水平在第3~7天达到峰值;肢体缺血再灌注初期,缺血时间越长,体内的血浆ET水平越高。给予止血带预处理,可显著降低大鼠体内血浆ET水平。
Objective To investigate the changes of plasma endothelin (ET) induced by tourniquet preconditioning in rats and the effect of ischemic preconditioning on it. Methods Sixty healthy SD rats weighing 150-200 g (male or female) with body weight of 150-200 g were randomly assigned to control group and experimental group with 30 in each. The control group without ischemic preconditioning, in the direct ischemia 2, 3, 4 h and the recovery of blood flow after the 1st, 3rd, 7th and 14th days to determine the plasma ET level; experimental group in the thigh to use the balloon to stop bleeding caused by limbs After ischemia, the tourniquet was relaxed for 10 min to form an ischemic preconditioning process, which was repeated 3 times. Blood was collected on days 2, 3, and 4 h after ischemia and on days 1, 3, 7 and 14 after resuscitation Plasma ET levels were measured. Results The levels of plasma ET in the same treatment time on the 1st, 3rd, 7th and 7th days of the two groups were compared, P <0.05 or <0.01; on the 14th day, the levels of plasma ET in the two groups were compared 3 h after ischemia (P <0.01). In the experimental group and the control group, plasma ET level increased first and then decreased gradually on the 1st, 3rd, 7th and 14th days, and reached its peak on the 3rd or 7th day (all P <0.05). The levels of plasma ET in the experimental group on the 1st, 3rd, 5th and 7th days were compared, P <0.05 or <0.01; P <0.05 or <0.01 on the 1st and 7th day in the control group. The levels of plasma ET in different experimental groups at 2, 3, and 4 hours after ischemia were lower than those in experimental group (P <0.01), and those in the control group at 2, 3 and 4 hours after ischemia were significantly lower than those of the control group (P <0.01). Conclusion In the limbs of rats with reperfusion injury, plasma ET levels peaked on day 3 to 7; in the early stage of limb ischemia / reperfusion, the longer the ischemic period, the higher the ET level in the body. Give tourniquet pretreatment, can significantly reduce plasma ET levels in rats.
