目的:观察夏枯草水提物对α-糖苷酶类活性、麦芽糖耐量试验的影响,研究其延缓正常ICR小鼠单糖吸收作用。方法:采用水煎法得到夏枯草水提物,按VanLoon′s碘法测定α-淀粉酶活性,pNPG法测定α-葡萄糖苷酶活性,葡萄糖氧化酶法测定正常ICR小鼠离体小肠α-麦芽糖酶活性;并测定夏枯草水提物对正常ICR小鼠麦芽糖耐量试验的动态血糖值的影响。结果:夏枯草水提物对α-淀粉酶、α-葡萄糖苷酶、离体小肠α-麦芽糖酶活性均有抑制作用,并呈量效关系;同时能提高其麦芽糖耐量,降低正常ICR小鼠餐后高血糖。结论:夏枯草水提物能延缓正常ICR小鼠单糖吸收,其机制可能与抑制肠道α-糖苷酶类有关。
OBJECTIVE: To observe the effects of P. vulgaris aqueous extracts on α-glucosidase activity and maltose tolerance test, and to investigate the effects of delaying the absorption of monosaccharides in normal ICR mice. Methods: The water extract of Prunella vulgaris was decocted, the α-amylase activity was measured by Van Loon’s iodine method, the α-glucosidase activity was measured by pNPG method, and the isolated small intestine α- was measured by glucose oxidase method in normal ICR mice. Maltase activity; and determination of the effect of Prunella vulgaris extract on the dynamic glucose level in maltose tolerance test in normal ICR mice. Results: Prunella vulgaris extracts inhibited α-amylase, α-glucosidase, and α-maltase activity in small intestine in vitro, and showed dose-effect relationship. At the same time, it could increase maltose tolerance and reduce normal ICR mice. Postprandial hyperglycemia. Conclusion: Prunella vulgaris extract can delay the absorption of monosaccharides in normal ICR mice. The mechanism may be related to the inhibition of intestinal α-glycosidases.
