目的 :应用高效毛细管电泳法对夏天无药材中的成分进行分离并测定其有效成分延胡索乙素的含量。方法 :运行缓冲液为CH3 OH 0 .6mol·L-1Tris溶液 (1∶1,用磷酸调pH =6 .0 ) ,另内含 10mmol·L-1庚烷磺酸钠 ;运行电压 2 0kV检测波长 2 0 0nm ;多潘立酮为内标。结果 :延胡索乙素在 0 .0 2 5~ 0 .2 5mg·ml-1范围内对内标峰面积之比成良好线性关系 (r=0 .9992 ) ,平均回收率为 98.4% ,RSD为 2 .6 % (n =5 )。结论 :实验结果表明 ,运用高效毛细管电泳法可将夏天无药材中的有效成分延胡索乙素与其它成分较好的分离 ,并准确测定其含量。
Objective : To apply the high performance capillary electrophoresis method to separate the components in summer without medicinal herbs and determine the content of its effective component tetrahydropalmatine. Method: The running buffer was CH3 OH 0 .6mol·L-1 Tris solution (1: 1, pH adjusted to pH=6.0 with phosphoric acid), and 10 mmol·L-1 sodium heptane sulphonate; running voltage 20 kV Wavelength 200 nm; domperidone as internal standard. Results: The ratio of the peak areas of internal standard in the range of 0.25-5.25 mg·ml-1 for Tetrahydropalmatine was linear (r=0.9992). The average recovery was 98.4%. The RSD was 2.6% (n = 5). Conclusion : The experimental results show that the use of high performance capillary electrophoresis can separate the effective component of tetrahydropalmatine in summer without herbs from other components and determine its content accurately.
