为了研究不同绿地土壤微生物活性特征,应用荧光素比色法和碱液吸收滴定法,对郑州市不同绿地的FDA(荧光素二乙酸酯)水解酶活性及微生物呼吸强度进行了分析,结果表明,不同绿地表层土壤FDA水解酶活性由高到低的顺序为:人工林地>人工草灌地>行道绿地(文化路)>行道绿地(诚信路)>隔离带绿地>人工草坪>人工灌木地>花卉绿地;人工林地、人工灌木地随土层深度增加FDA水解酶活性、土壤微生物呼吸强度呈下降趋势;FDA水解酶活性与微生物呼吸强度相关系数(r)为0.939 7,这表明FDA水解酶活性与土壤微生物呼吸强度呈正相关,两者均能反映土壤微生物活性,且荧光素比色法优势在于简单、快速、灵敏,这种方法测定总微生物的活性具有广泛性,故FDA水解酶活性更能反映土壤总微生物的活性。
In order to study the characteristics of soil microbial activity in different greenbelt, the activity of fluorescein diacetate hydrolase and the respiration intensity of microorganisms in different greenbaths in Zhengzhou City were analyzed by using fluorescein colorimetric method and lye titration. The activity of FDA hydrolase and the respiration intensity of soil microorganism decreased. The correlation coefficient (r) between FDA hydrolase activity and respiration rate of microorganism was 0.939 7, indicating that the activity of FDA hydrolase And soil respiration intensity was positively correlated, both of which can reflect the activity of soil microorganisms, and the advantages of the fluorescence method is simple, rapid and sensitive, this method of determination of total microbial activity is broad, so the FDA hydrolase activity more Reflects the activity of total soil microorganisms.
