目的 研究HLA DQA1基因与广西地区壮族、汉族 2型糖尿病 (DM )的关联。方法 采用聚合酶链反应 序列特异性引物 (PCR SSP)技术 ,分别对广西 67例壮族和 72例汉族 2型DM及 69例壮族、65例汉族正常对照的HLA DQA1位点进行基因分型。结果 壮族 2型DM组DQA1 0 3 0 1、DQA1 0 40 1等位基因频率显著高于壮族对照组 (P <0 .0 0 1,RR =4.69和P <0 .0 0 1,RR =7.5 5 ) ,汉族 2型DM组DQA1 0 10 4等位基因频率明显高于正常汉族对照组 (P =0 .0 0 1,RR =6.5 1) ;DQA1 0 60 1在壮族 2型DM组明显低于对照组 (P <0 .0 0 1,RR =0 .14 ) ,DQA1 0 40 1等位基因频率在汉族 2型DM组明显低于汉族对照组 (P <0 .0 0 1,RR =0 .2 9)。结论 DQA1 0 40 1、DQA1 0 3 0 1可能是广西壮族 2型DM的易感基因 ,而DQA1 0 10 4可能为汉族 2型DM的易感基因 ;DQA1 0 60 1可能是壮族 2型DM的保护基因 ,DQA1 0 40 1则可能是汉族 2型DM的保护基因
Objective To study the association between HLA DQA1 gene and type 2 diabetes mellitus (DM) in Zhuang and Han in Guangxi Province. Methods Polymerase chain reaction sequence specific primers (PCR SSP) were used to genotype HLA DQA1 loci in 67 cases of Zhuang nationality in Guangxi province, 72 cases of Han type 2 DM group and 69 cases of Zhuang nationality in Han nationality. Results The frequencies of DQA1 0 3 0 1 and DQA 1 0 40 1 alleles in Zhuang type 2 DM group were significantly higher than those in Zhuang control group (P <0. 001, RR = 4.69 and P <0.01, RR = 7.5 5). The frequency of DQA1 0 10 4 allele in Han type 2 diabetes mellitus group was significantly higher than that in normal Han controls (P = 0.001, RR = 6.51). DQA1 0 60 1 was significantly lower in Zhuang 2 DM group In control group (P <0.01, RR = 0.14), the frequency of DQA1 0 40 1 allele in Han-type DM group was significantly lower than that in Han control group (P <0.01, RR = 0 .2 9). Conclusion DQA1 0 40 1 and DQA1 0 3 0 1 may be susceptible genes of type 2 DM in Guangxi Zhuang Autonomous Region, while DQA1 0 10 4 may be susceptible genes of type 2 DM in Han Nationality. DQA1 0 60 1 may be a susceptible gene of Zhuang type 2 DM The protective gene, DQA1401, may be a protective gene for Han type 2 DM
