目的:研究高糖刺激肾小球系膜细胞后,凋亡相关基因Bcl-2、Smac、Fas的表达情况。方法:将体外培养的大鼠肾小球系膜细胞分为正常对照组(糖浓度为5.6 mmol/L)和高糖组(糖浓度为30 mmol/L),采用四甲基偶氮唑蓝(MTT)比色法检测不同作用时间(12 h、24 h、36 h、48 h)下,系膜细胞的增殖状况,且在各时间点,采用免疫荧光染色法观察Bcl-2、Smac、Fas的表达情况并测定其荧光强度。结果:MTT比色法检测结果显示高糖组较对照组系膜细胞增殖明显,与作用时间呈正相关。同时,免疫
