目的 评价外源性PTEN抑癌基因对人高转移性粘液表皮样癌细胞系M 3SP2药物敏感性的影响。方法 用脂质体介导法将PTEN抑癌基因导入M 3SP2细胞系 ,以空载体转染细胞为对照 ,然后应用MTT比色法测定两种癌细胞对 10种常用抗癌药物敏感性。结果 与对照细胞比较 ,PTEN基因转染细胞对 6种抗癌药物甲氨喋呤、重组人干扰素 -γ、丝裂霉素、多西环素、平阳霉素和氟脲嘧啶的相对抗肿瘤活性呈不同程度地增强 ,药物增敏比增加了 1.5~ 5 2 .1倍。结论 外源性野生型PTEN抑癌基因能增强粘液表皮样癌细胞对常用抗癌药物的敏感性。
Objective To evaluate the effect of exogenous PTEN oncogene sensitivity to M 3SP2 in human highly metastatic mucoepidermoid carcinoma cell line. Methods PTEN tumor suppressor gene was transfected into MSP2 cell line by liposome-mediated method. The empty vector transfected cells were used as control. MTT assay was used to determine the sensitivity of the two kinds of cancer cells to 10 commonly used anti-cancer drugs. Results Compared with the control cells, PTEN gene transfected cells showed significant anti-tumor effect on six anti-cancer drugs methotrexate, recombinant human interferon-γ, mitomycin, doxycycline, bleomycin and fluorouracil Activity was increased to varying degrees, the drug sensitization increased by 1.5 to 52.1 times. Conclusion The exogenous wild-type PTEN tumor suppressor gene can enhance the sensitivity of mucoepidermoid carcinoma cells to commonly used anti-cancer drugs.
