目的 观察维甲酸(RA)纳米粒对兔视网膜有无毒性作用。方法 30只青紫蓝兔随机分为5组:正常组,RA 5μg/ml组,RA 10μg/ml组,RA 20μg/ml组及对照组,分别于玻璃体腔注药后7、14及28 d用检眼镜观察眼底变化,然后摘除眼球,进行组织学观察,视网膜神经节细胞(RGCs)计数并测量视网膜内层厚度。结果 7 d时,RA 20μg/ml组出现 RGCs的空泡样变,但在14 d时恢复正常。各组之间RGCs数目及视网膜内层厚度无差异。结论 ≤20μg/ml RA注入玻璃体腔对于视网膜是安全的,没有对视网膜产生远期毒性。
Objective To observe whether retinoic acid (RA) nanoparticles have toxic effects on rabbit retina. Methods Thirty black and blue rabbits were randomly divided into 5 groups: normal group, RA 5μg / ml group, RA 10μg / ml group, RA 20μg / ml group and control group. The ophthalmoscope was used to observe the fundus changes. The eyeballs were then removed for histological observation. Retinal ganglion cells (RGCs) were counted and the thickness of the retina was measured. Results At 7 days, the vacuolar degeneration of RGCs appeared in RA 20μg / ml group, but returned to normal after 14 days. There was no difference in the number of RGCs and the thickness of retina between groups. Conclusion The injection of 20μg / ml RA into the vitreous cavity is safe for the retina without long-term toxicity to the retina.
