为研究人参水提物(WEG)对SW480细胞体外增殖的抑制作用,采用倒置显微镜、MTT法观测SW480细胞的存活情况,并通过原子力显微镜获取各试验组细胞在人参水提物作用下细胞膜的平面/三维超微结构。结果表明:人参水提物能显著抑制SW480细胞增殖,其凋亡率与作用时间、剂量存在依赖关系(P<0.05),随着作用时间延长或剂量加大,对SW480细胞抑制作用呈增加趋势。原子力显微图像表明,正常SW480细胞呈圆形、细胞饱满,表面较光滑,而WEG处理组存活细胞皱缩,细胞表面的粗糙度较大,细胞膜存在一定的损伤。这说明WEG能显著抑制SW480细胞体外增殖,可能与WEG对SW480细胞膜的损伤有关。
In order to study the inhibitory effect of ginseng water extract (WEG) on the proliferation of SW480 cells in vitro, the survival of SW480 cells was observed by MTT method and the cell membrane surface of each experimental group was obtained by atomic force microscopy Three-dimensional ultrastructure. The results showed that the water extract of Panax ginseng could significantly inhibit the proliferation of SW480 cells. The apoptosis rate of SW480 cells was dependent on the time and dose (P <0.05). The inhibition of SW480 cells increased with time or dose . Atomic force microscopy images showed that the normal SW480 cells were round, full of cells, and the surface was smooth. However, the surviving cells of WEG treated group were shrunk, and the roughness of cell surface was larger and cell membrane was damaged to some extent. This shows that WEG can significantly inhibit the proliferation of SW480 cells in vitro, which may be related to WEG on SW480 cell membrane damage.
