目的研究氯化两面针碱体外促进两种鼻咽癌细胞株凋亡的作用。方法应用MTT法检测细胞的增殖活性,DNA ladder检测、细胞形态学观察、酶联免疫法进行检测分析。结果氯化两面针碱对两种鼻咽癌7111,Ecv2细胞株的IC50分别为1.238±0.09、1.641±0.331μg.ml-1。在其作用下,两种鼻咽癌细胞株出现典型的凋亡特征,琼脂糖电泳出现典型的DNA梯带,细胞形态学表现出细胞核裂解、染色质聚集、核碎裂,胞浆浓缩、有空泡形成,ELISA法检测样本活性后发现药物作用后细胞中Caspase-3含量明显高于对照组(P<0.05)。结论氯化两面针碱有促进两种鼻咽癌7111,Ecv2细胞株凋亡的作用。
Objective To study the effect of nitidine chloride on the apoptosis of two nasopharyngeal carcinoma cell lines in vitro. Methods MTT assay was used to detect the cell proliferation activity, DNA ladder test, cell morphology observation and enzyme-linked immunosorbent assay. Results The amphotropic nitidine chloride had an IC50 of 1.238 ± 0.09,1.641 ± 0.331μg.ml-1 for the two NPC cell lines 7111 and Ecv2, respectively. Under the action of these two nasopharyngeal carcinoma cell lines, typical apoptotic features were observed. Typical DNA ladder appeared on agarose gel electrophoresis. Cell morphology showed nuclear lysis, chromatin aggregation, nuclear fragmentation and cytoplasm concentration. There were The formation of vacuoles and the activity of Caspase-3 in the cells detected by ELISA were significantly higher than those in the control group (P <0.05). CONCLUSION Chlorpheniramine can promote the apoptosis of the two nasopharyngeal carcinoma cell lines 7111 and Ecv2.
