Rubisco是光合作用和光呼吸的关键酶,目前已成为高等植物中研究最深入的酶之一,而在低等藻类中关于该酶的相关研究却很少。以来自浙江和福建的6个坛紫菜(Porphyra haitanensis)无性系为研究对象,以江苏条斑紫菜(P.yezoensis)为参照,克隆了其Rubisco小亚基(rbcS)及大小亚基基因间隔区共516 bp的序列。序列分析结果表明,其中4个坛紫菜系(Hza、Hzf、Hpt、Hzz)的基因序列完全相同(以其作为标准),坛紫菜Hzb和Hzc与其均只有1个碱基差异,而条斑紫菜Yn55与其差别碱基为46个。将本实验的7条紫菜序列与GenBank数据库收录的17条紫菜Rubisco序列一起做系统分析,结果表明坛紫菜和条斑紫菜都分别归到了各自的类群中。以上结果说明,紫菜rbcS和rbcL-rbcS基因间隔区序列的同源性较高,可用于种以上水平的系统分析。
Rubisco is one of the most studied enzymes in photosynthesis and photorespiration and has become one of the most studied enzymes in higher plants. However, few studies have been done on this enzyme in lower algae. The clones of six Porphyra haitanensis clones from Zhejiang and Fujian Provinces were selected as the research objects. The Rubisco small subunit (rbcS) and the size subunit gene spacer region were cloned from P. yezoensis A total of 516 bp of sequence. The results of sequence analysis showed that the sequences of Hza, Hzf, Hpt and Hzz were exactly the same (as standard), and the frequencies of Hzb and Hzc were only 1 base difference, The difference between Yn55 and base 46 is 46. The seven seaweed sequences in this experiment were systematically analyzed with 17 Rubisco sequences from GenBank. The results showed that the algae and the seaweed belonged to their respective taxa. The above results showed that the homology between the laver rbcS and rbcL-rbcS intergenic spacer sequences is high and can be used for systematic analysis above the species level.
