目的观察过氧化物酶体增殖物激活受体(PPAR)α、γ配体对泡沫细胞内胆固醇酯、ox-LDL 和三磷酸腺苷结合盒转运体 A1(ABCA1)、小凹蛋白-1(caveolin-1)表达的影响。方法用TBARS 法检测细胞内 MDA。用油红 O 染色法检测泡沫细胞的形成。用荧光分光光度法检测细胞内总胆固醇、游离胆固醇和胆固醇酯含量。ELISA 法检测泡沫细胞内 ox-LDL。用 RT-PCR 和 Westernblot 检测 ABCA1、caveolin-1 mRNA 与蛋白的表达。结果单核细胞经佛波醇酯和 ox-LDL 孵育后,有大量泡沫细胞形成,细胞内积聚的总胆固醇、游离胆固醇、胆固醇酯及脂质过氧化产物、ox-LDL 均明显增多(P<0.05),而 ABCA1、caveolin-1蛋白、mRNA 水平明显减少(P<0.05),PPARα、γ配体干预组显著缓解上述变化,细胞内胆固醇酯显著减少,ABCA1、caveolin-1 mRNA 和蛋白表达明显增加(P<0.05)。结论 PPARα、γ配体可能通过增强 ABCA1、caveolin-1的表达和降低细胞内胆固醇聚积发挥抗动脉粥样硬化的作用。
Objective To observe the effects of peroxisome proliferator - activated receptor (PPAR) α and γ ligand on the intracellular cholesterol ester, ox - LDL and adenosine triphosphate binding cassette transporter A1 (ABCA1), caveolin - 1 ) Expression. Methods The intracellular MDA was detected by TBARS method. The formation of foam cells was examined by oil red O staining. The contents of total cholesterol, free cholesterol and cholesterol ester in the cells were detected by fluorescence spectrophotometry. Ox-LDL in foam cells was detected by ELISA. The expression of ABCA1 and caveolin-1 mRNA and protein were detected by RT-PCR and Western blot. Results After monocytes were incubated with phorbol esters and ox-LDL, a large number of foam cells were formed. Total cholesterol, free cholesterol, cholesteryl ester and lipid peroxidation products and ox-LDL in cells were significantly increased (P < 0.05). The levels of ABCA1 and caveolin-1 mRNA and protein were significantly decreased (P <0.05). The PPARα and γ ligand intervention groups significantly alleviated the above changes, the intracellular cholesterol esters decreased significantly, and the expressions of ABCA1 and caveolin-1 mRNA and protein Increase (P <0.05). Conclusion PPARα and γ ligands may play an anti-atherosclerotic role by enhancing the expression of ABCA1 and caveolin-1 and decreasing the intracellular cholesterol accumulation.
