融合蛋白的设计中,经常需要用链接肽将不同功能的蛋白进行连接融合。连接肽的设计和选择对融合蛋白的表达、稳定性及功能活性至关重要。文章将常见的两种连接肽分为柔性(Flexibility)和刚性(Rigidity)进行比较说明。其中柔性连接肽即成柔软线性的一类易弯折的氨基酸序列,常见为(GGGGS)n重复序列(n=3~4);刚性连接肽即成稳固螺旋结构的一类长度固定的氨基酸序列,常见为(EAAAK)n重复序列(n=2~5)。设计连接肽时应考虑长度、氨基酸成分、亲水性、构象、柔韧性及电荷等因素。连接肽中的氨基酸组成对柔性连接肽影响较大,而氨基酸排列影响着刚性连接肽的结构。文章结合近年来融合蛋白设计中两种最常用连接肽的相关报道进行阐述,为连接肽的研究提供参考。
In the design of fusion proteins, it is often necessary to link and combine proteins of different functions with the linked peptide. The design and selection of the linker peptide is crucial for the expression, stability and functional activity of the fusion protein. The article will be common to the two peptides into the connection flexibility (Flexibility) and rigidity (Rigidity) for comparison. The flexible linker peptide is a kind of pliable amino acid sequence of soft linearity, commonly known as (GGGGS) n repeats (n = 3-4); the rigid linker peptide is a kind of fixed length amino acid sequence of stable helical structure , Common (EAAAK) n repeat (n = 2 ~ 5). The linker should be designed with respect to length, amino acid composition, hydrophilicity, conformation, flexibility and charge. The amino acid composition of the linker peptide has a great effect on the flexible linker peptide, and the amino acid alignment affects the structure of the rigid linker peptide. In this paper, we report the related reports of the two most commonly used linker peptides in fusion protein design in recent years, and provide references for the study of linker peptides.
