目的分离纯化虎杖发酵液中的白藜芦醇。方法利用大孔树脂结合聚酰胺柱色谱从虎杖发酵液的提取液中纯化白藜芦醇,并研究了大孔树脂和聚酰胺对白藜芦醇的吸附动力学。结果大孔树脂和聚酰胺对白藜芦醇的表观静态吸附量分别为11.70 mg/g和57.9 mg/g,表观动态吸附量分别为12.47 mg/g和58.98 mg/g,吸附行为符合Langmuir动力学。利用大孔树脂结合聚酰胺两步柱色谱纯化提取液中的白藜芦醇,使其纯度达到95.8%,总收率达69.8%。结论此方法操作简单,成本低廉,白藜芦醇的纯度和收率高,有工业化应用前景。
Objective To isolate and purify resveratrol from the fermentation broth of Polygonum cuspidatum. Methods Resveratrol was purified from the extract of Polygonum cuspidatum by macroporous resin and polyamide column chromatography. The adsorption kinetics of resveratrol by macroporous resin and polyamide was studied. Results The apparent static adsorption capacities of resorcinol with macroporous resin and polyamide were 11.70 mg / g and 57.9 mg / g, respectively. The apparent dynamic adsorption capacities were 12.47 mg / g and 58.98 mg / g, respectively. The adsorption behavior was consistent with Langmuir dynamics. Resveratrol in the extract was purified by macroporous resin and polyamide two-step column chromatography, the purity was 95.8% and the total yield was 69.8%. Conclusion This method is simple, low cost, high purity and yield of resveratrol, and has industrial application prospect.
