分别对不同继代次数(3~90代)‘富士’、‘金冠’、‘乔纳金’苹果组培苗进行POX、EST、AMY同工酶分析。结果表明,同一品种不同继代次数处理间均有一致的酶带谱型。利用超薄层等电聚焦电泳根据POX酶蛋白pI的差异可区分金冠和乔纳金两品种,同一品种不同继代次数的组培苗之间pI相同,组培过程未对POX酶蛋白pI产生影响。利用25个随机引物对不同继代次数的苹果组培继代苗进行了RAPD分析,其特征带型表现一致,未发现变异。说明苹果茎尖离体继代培养90次以内具有较高的遗传稳定性。
The isozymes analysis of POX, EST and AMY were conducted on the tissue culture shoots of ’Fuji’, ’Golden Delicious’ and ’Jonathan’ apple from different generations (from 3 to 90 generations). The results showed that there was a consistent pattern of enzyme bands between different subcultures of the same variety. Using ultrathin layer isoelectric focusing electrophoresis, the pI of POX protein can be distinguished according to the difference of the pI of POX enzyme, and the pI between the two cultivars with the same subculture number and the subculture duration of the same variety can be distinguished. influences.
