目的建立类似于人类多发性肌炎(PM)的豚鼠模型,观察天龙液对此模型的治疗作用。方法于2003年11月至2004年5月对吉林大学第一医院应用兔肌肉匀浆加完全佛氏佐剂免疫剂,注射豚鼠制成实验性自身免疫性肌炎(EAM)动物模型。并将EAM豚鼠随机分为4组:A组给予小剂量天龙液[1250mg/(kg.d)]治疗,B组给予大剂量天龙液[3750mg/(kg.d)]治疗,C组给予泼尼松[3mg/(kg.d)]治疗,D组作为空白对照,观察比较临床表现、血清肌酶、肌电图及肌肉病理改变。结果A、B、C3组的临床表现、血清肌酶、肌电图、肌肉病理改变明显好转(各组治疗前后比较P<0.05)。D组无好转。结论本实验所建立的EAM豚鼠模型与人类PM相似,天龙液对EAM豚鼠有治疗作用,为今后临床应用天龙治疗PM提供了依据。
Objective To establish a guinea pig model similar to human polymyositis (PM) and observe the therapeutic effect of Tianlong liquid on this model. Methods The animal model of experimental autoimmune myositis (EAM) was made by injecting guinea pigs into the first hospital of Jilin University from November 2003 to May 2004 using rabbit muscle homogenate and complete Freund’s adjuvant. EAM guinea pigs were randomly divided into 4 groups: Group A was given a small dose of Tianlongye [1250mg/(kg.d)], Group B was given a large dose of Tianlongye [3750mg/(kg.d)], and Group C was given a splash. Nisson [3mg/(kg.d)] treatment, D group as a blank control, observation and comparison of clinical manifestations, serum muscle enzymes, EMG and muscle pathological changes. Results The clinical manifestations, serum muscle enzymes, electromyogram, and muscle pathology in group A, B, and C3 were significantly improved (P<0.05 for each group before and after treatment). Group D did not improve. Conclusion The EAM guinea pig model established in this experiment is similar to human PM. Tianlong liquid has therapeutic effects on EAM guinea pigs, providing a basis for clinical application of Tianlong in PM treatment.
