作者运用生物素标记的山羊抗人巨噬细胞甘露糖受体 (GoatAnti humanMacrophageMannoseRecep tor ,AMMR)IgG对人获能精子膜上的甘露糖结合蛋白进行定位 ,经激光共聚焦显微镜分析结果显示 :AMMRIgG组阳性率显著高于运用正常山羊 (NormalGoat,NG)IgG的对照组 :AMMRIgG组的阳性率大约在 70 % (6 9.41% )左右 ,而NGIgG组小于 10 % (9.2 4% )。AMMRIgG组中 ,荧光结合部位主要在精子的顶体后区、赤道区及颈部 ,而NGIgG对照组中除少量精子显微弱的荧光外 ,绝大多数呈阴性结果。精子膜上甘露糖结合蛋白的定位研究可能对受精过程中精卵识别机制的了解具有重要意义。
The authors used Goat Anti human Macrophage MannoseRecep tor (AMMR) IgG to locate the mannose binding protein on human-accessible sperm membrane. Confocal laser scanning microscopy results showed that the AMMRIgG group The positive rate was significantly higher than that of normal goat IgG: the positive rate of AMMRIgG was about 70% (6 9.41%), while that of NGIgG was less than 10% (9.2 4%). In the AMMRIgG group, most of the fluorescence binding sites were found in the posterior acrosome region, the equatorial region and the neck of the sperm, while the NGIgG control group showed the most negative results except a small amount of sperm fluorescence. The localization of mannose-binding proteins in sperm membrane may be of great significance for the understanding of sperm-egg recognition mechanism during fertilization.
