目的观察雌激素处理对成年雄性小鼠黑质多巴胺神经元数量的影响及可能的调控机制。方法选用雄性C57/BL6小鼠10只,分成对照组、雌激素组(E2),采用免疫组化检测雌激素处理对小鼠黑质多巴胺神经元与calb ind in阳性神经元数量的改变;采用RT-PCR分析雌激素处理对小鼠中脑内多巴胺转运蛋白(DAT)、胰岛素样生长因子1受体(IGF-1R)与脑源性神经营养因子(BDNF)mRNA水平的影响。结果雌激素处理可以增加小鼠黑质多巴胺神经元数量[E2组(119.7±2.9)个vs对照组(93.5±5.1)个,P=0.004],并增加小鼠中脑内DAT的表达水平(P<0.05),而对calb ind in阳性神经元数量无影响[E2组(88.0±2.0)个vs对照组(86.2±1.5)个]。雌激素处理同时可以上调小鼠中脑内IGF-1R与BDNFmRNA水平(P<0.05)。结论雌激素通过调节IGF-1R与BDNF的表达增加小鼠中脑内多巴胺神经元数量。
Objective To observe the effect of estrogen on the number of dopaminergic neurons in substantia nigra of adult male mice and the possible regulatory mechanism. Methods Ten male C57 / BL6 mice were divided into control group and estrogen group (E2). Immunohistochemistry was used to detect the changes of the numbers of substantia nigra dopamine neurons and calb ind in positive neurons in mice. The effect of estrogen on mRNA levels of dopamine transporter (DAT), insulin-like growth factor 1 receptor (IGF-1R) and brain-derived neurotrophic factor (BDNF) in the midbrain of mice was analyzed by RT-PCR. Results Estrogen treatment increased the number of dopaminergic neurons in substantia nigra of mice [119.7 ± 2.9 in E2 group vs 93.5 ± 5.1 in control group (P = 0.004), and increased the expression of DAT in the midbrain of mice P <0.05), but had no effect on the number of calb ind in positive neurons (88.0 ± 2.0 in E2 group vs 86.2 ± 1.5 in control group). Estrogen treatment also increased IGF-1R and BDNF mRNA levels in the midbrain of mice (P <0.05). Conclusion Estrogen increases the number of dopaminergic neurons in the midbrain of mice by regulating the expression of IGF-1R and BDNF.
