在培养食用菌母种时,人们通常沿用玻璃试管琼脂斜面培育方法,管口用棉塞封闭.这种方法有不少优点,但也有不足之处,如PDA所培养的母种基质易干涸、液化,存放过程中易遭杂菌污染,菌丝易老化,转接次数多.为此笔者从1991年起开始试用盐水瓶培育平菇母种,取得理想的效果.现将具体方法介绍如下:1 准备材料①取100mL的盐水瓶,清洗后用0.1%新洁尔灭液冲洗,橡皮塞也同时冲洗.②取2mm口径的硬质塑料管6cm,在酒精灯上于2.5cm处弯成90°直角,在短段用刀削成斜面.③培养料为阔叶木屑和木粒(3~5mm)各半,玉米粉10%,石膏1%,石灰、酵母粉各2%,磷酸二氢钾、硫酸镁各0.1%,水65%.
