目的研究低剂量脉冲超声波(low-intensity pulsed ultrasound,LIPUS)对体外培养细胞的生物学作用。方法以LIPUS作用于人皮肤成纤维细胞0~40 min,采用MTT比色法、酶谱法和Western blot法观察细胞增殖效应。以LIPUS作用于人皮肤成纤维细胞和人胰腺癌细胞(Mia Pa Ca-2)0~100 min,用Annexin V法检测凋亡效应。结果在0~40 min LIPUS刺激下,细胞活性、细胞所分泌的活性介质等增加,细胞内蛋白磷酸化水平升高;LIPUS刺激超过60 min后,凋亡细胞数增加,且LIPUS刺激达100 min时,凋亡细胞数增加更加明显。结论短时间LIPUS刺激能促进细胞增殖,超过一定时间则会抑制细胞生长,甚至引起细胞凋亡。
Objective To study the biological effects of low-dose pulsed ultrasound (LIPUS) on cultured cells in vitro. Methods Human dermal fibroblasts were treated with LIPUS for 0-40 min. MTT colorimetric assay, zymography and Western blot were used to observe the cell proliferation. LIPUS was applied to human skin fibroblasts and human pancreatic cancer cells (Mia Pa Ca-2) for 0-100 min, and the apoptosis effect was detected by Annexin V method. Results After LIPUS stimulation for 0-40 min, the activity of cells and the active mediators secreted by the cells increased and the phosphorylation of intracellular protein increased. After 60 min stimulation with LIPUS, the number of apoptotic cells increased and LIPUS stimulation for 100 min When the number of apoptotic cells increased more significantly. Conclusion LIPUS stimulation can promote cell proliferation in a short time, and over a certain period of time can inhibit cell growth and even cause apoptosis.
