目的探讨硫酸镍(NiSO4)对大鼠肾细胞凋亡相关基因C-kit、Caspase-3和Bax蛋白表达及氧化应激水平的影响。方法选择健康Wistar雄性大鼠32只,随机分为4组,即生理盐水(NS)组,NiSO4 1.25、2.5和5.0 mg/kg组,腹腔注射染毒,1次/d,连续30 d。采用Western Bloting技术检测大鼠肾细胞凋亡相关基因C-kit、Caspase-3和Bax的蛋白表达水平;分光光度法测定大鼠肾组织匀浆超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、丙二醛(MDA)和过氧化氢(H2O2)。结果 NiSO4 2.5 mg/kg及其以上组可引起C-Kit蛋白表达水平升高,而Caspase-3和Bax蛋白表达水平分别为5.0和2.5 mg/kg组表达上调,并使SOD、GSH-Px和CAT酶活力降低,H2O2含量上调,与对照组比较,差异有统计学意义(P<0.05)。结论在该实验条件下,NiSO4大鼠肾损伤可能与c-kit、Caspase-3、Bax蛋白表达和氧化应激效应异常改变有关。
Objective To investigate the effects of NiSO4 on expression of C-kit, Caspase-3 and Bax proteins and oxidative stress in rat renal cell. Methods Thirty-two healthy Wistar male rats were randomly divided into 4 groups: normal saline (NS) group, NiSO4 1.25, 2.5 and 5.0 mg / kg group, intraperitoneal injection once a day for 30 days. Western Bloting was used to detect the protein expression of C-kit, Caspase-3 and Bax in rat renal cell apoptosis. Spectrophotometry was used to determine the content of superoxide dismutase (SOD), glutathione Peroxidase (GSH-Px), catalase (CAT), malondialdehyde (MDA) and hydrogen peroxide (H2O2). Results NiSO4 2.5 mg / kg and above resulted in an increase in the expression of C-Kit protein, while the expressions of Caspase-3 and Bax protein were up-regulated in 5.0 and 2.5 mg / kg groups, respectively. SOD, GSH-Px and CAT activity decreased, H2O2 content increased, compared with the control group, the difference was statistically significant (P <0.05). Conclusion Under the experimental conditions, the renal injury of NiSO4 rats may be related to the abnormal expression of c-kit, Caspase-3, Bax protein and oxidative stress.
