目的筛选可产生多种生物活性物质的噬几丁质属Chitinophaga菌株。方法采用VY/4平板法从兔粪、羊粪、泥土、朽木等样品中筛选目的菌株;用滤纸片法检测抗菌活性;用纤维蛋白平板法检测溶栓活性;用纤维蛋白原平板法检测抗凝活性;用形态学特征、理化性质、16S rRNA序列同源性鉴定目的菌株。结果从朽木样品中分离到一株可同时产生广谱抗生素和抗血栓活性物质的ZLX3菌株,其分泌的胞外液具明显的广谱抗菌作用和溶栓抗凝活性;结合形态学、理化性质、16S rRNA序列同源性及进化树分析,发现ZLX3菌株属于Chitinophaga属,将其命名为Chitinophaga sp.ZLX3。结论 ZLX3菌株可产生广谱抗生素和抗凝溶栓活性物质。
Objective To screen Chitinophaga strains which can produce a variety of bioactive substances. Methods VY / 4 plate method was used to screen the target strains from rabbit feces, sheep manure, mud, dead wood and other samples. The antibacterial activity was tested by filter paper method. The thrombolytic activity was tested by fibrin plate method. Coagulation activity; the morphological characteristics, physical and chemical properties, 16S rRNA sequence homology identification of the target strain. Results A strain of ZLX3 which can produce broad-spectrum antibiotics and antithrombotic active substances was isolated from turmeric samples. The secreted extracellular fluid has obvious broad-spectrum antibacterial activity and thrombolytic anticoagulant activity. The morphological and physicochemical properties , 16S rRNA sequence homology and phylogenetic tree analysis. It was found that ZLX3 belongs to Chitinophaga genus and named it Chitinophaga sp.ZLX3. Conclusion ZLX3 strains can produce broad-spectrum antibiotics and anticoagulant thrombolytic active substances.
