目的探索人胚干细胞来源的成纤维细胞(EBF)作为纳米银颗粒细胞毒性评价模型的可行性,旨在为纳米材料生物安全性评价提供新的细胞模型。方法将纳米银与EBF细胞共孵育,采用相差显微镜、CCK-8法、流式细胞术和透射电子显微镜分别对细胞形态、细胞增殖、细胞周期及细胞摄取进行研究。结果纳米银颗粒对EBF活性的影响是浓度和时间依赖性的,纳米银可以进入细胞内并引起细胞周期阻滞。结论 EBF可以作为一种新的纳米银颗粒生物安全性评价模型,由于其来源的特殊性,可进一步模拟人体正常细胞的反应。
Objective To explore the feasibility of human embryonic stem cell-derived fibroblasts (EBF) as a model for assessing the cytotoxicity of nano-silver particles, aiming to provide a new cell model for the biosafety evaluation of nano-materials. Methods Silver nanoparticles were incubated with EBF cells, and the morphology, cell proliferation, cell cycle and cell uptake were studied by phase contrast microscopy, CCK-8 assay, flow cytometry and transmission electron microscopy. Results The effect of nanosilver particles on EBF activity was concentration-dependent and time-dependent. Nanosilver could enter into cells and cause cell cycle arrest. Conclusion EBF can be used as a new bio-safety evaluation model of nano-silver particles. Because of its special source, EBF can further simulate the reaction of human normal cells.
