采用水提醇沉法,通过单因素和正交试验,对提取紫红牛肝菌粗多糖的最佳方法进行了筛选,并采用DPPH法对其活力进行了测定。结果表明:提取紫红牛肝菌粗多糖最佳方法为微波法,最佳去色素条件为活性炭0.2 g,水浴时间1.5 h,水浴温度20℃。最佳工艺条件为:微波时间2 min、料液比1:30、提取次数一次,粗多糖的得率为18.84 mg/g。当紫红牛肝菌粗多糖溶液浓度为1.3 mg/mL时,抗氧化活性为92.56%,且提取液中粗多糖的含量与抗氧化活性成正比,说明紫红牛肝菌粗多糖具一定的抗氧化活性。
Water extraction alcohol precipitation method, through single factor and orthogonal test, the best method of extracting crude polysaccharide from Bruguiera purpura was screened, and its activity was determined by DPPH method. The results showed that the best method for extracting crude polysaccharides from Bruguiera sexangula was microwave method. The optimum conditions for the pigment removal were activated carbon 0.2 g, water bath time 1.5 h and water bath temperature 20 ℃. The optimum conditions were as follows: the microwave time was 2 min, the solid-liquid ratio was 1:30, the extraction times were once, and the crude polysaccharide yield was 18.84 mg / g. Antioxidant activity was 92.56% when the concentration of crude polysaccharides of Bruguiera sexangula was 1.3 mg / mL, and the content of crude polysaccharides in the extract was in direct proportion to the antioxidant activity, indicating that crude polysaccharides of Bruguiera sexangula with certain anti-oxidation active.
