目的 :高效液相色谱法 (HPLC)测定家兔体内布比卡因的血药浓度。方法 :色谱柱为Nova PakC18柱。流动相为甲醇 0 0 5mol·L-1醋酸缓冲液 35∶6 5 (V/V) ,紫外检测波长 2 6 5nm ,柱温 (40± 0 5 )℃。结果 :布比卡因的保留时间为 9 2 0min ,与血浆峰分离良好 ,无内源性物质的干扰。在 5~ 10 0 μg·mL-1浓度范围内线性关系良好 ,r =0 994 0 (n =7)。最低检测浓度 2 5 μg·mL-1。结论 :本方法适用于布比卡因的药动学研究。
Objective: To determine the plasma concentration of bupivacaine in rabbits by high performance liquid chromatography (HPLC). Method: The column was a Nova Pak C18 column. The mobile phase consisted of methanol 0 0 5mol·L-1 acetate buffer 35:65 (V / V), ultraviolet detection wavelength 265nm and column temperature (40 ± 0 5) ℃. Results: The retention time of bupivacaine was 920 min, which was well separated from the plasma peak without the interference of endogenous substances. There was a good linear relationship in the range of 5 ~ 100 μg · mL-1, r = 0 994 0 (n = 7). The lowest detection concentration of 25 μg · mL-1. Conclusion: This method is suitable for pharmacokinetics of bupivacaine.
