目的探讨模拟失重对胃癌SGC-7901细胞和胃黏膜上皮HFE-145细胞增殖和周期的影响。方法采用回转器模拟失重,两种细胞各分为两组,回转组和1G对照组,实验时间共72h。采用免疫组化法观察两种细胞增殖核抗原(proliferating cell nuclear antigen,PCNA)表达的变化,应用流式细胞仪测定细胞周期。结果与对照组相比,回转组SGC-7901细胞48h及72hPCNA抗体阳性率明显减少(P<0.05及P<0.01),G0+G1期细胞显著增多(P<0.01及P<0.05);12h、24h及36h PCNA抗体阳性细胞的比率无显著性差异。与对照组相比,回转组HFE-145细胞12hPCNA抗体阳性率明显减少(P<0.05),G0+G1期细胞显著增多(P<0.05)。结论回转器模拟失重使胃癌SGC-7901细胞48h及72h时出现细胞周期阻滞、增殖抑制。胃黏膜上皮HFE-145细胞在12h时出现上述变化。
Objective To investigate the effects of simulated weightlessness on the proliferation and cell cycle of gastric cancer cell line SGC-7901 and gastric epithelial HFE-145 cells. Methods The rotator was used to simulate weightlessness. The two kinds of cells were divided into two groups, swivel group and 1G control group. The experiment time was 72h. Immunohistochemistry was used to observe the changes of proliferating cell nuclear antigen (PCNA) expression. The cell cycle was determined by flow cytometry. Results Compared with the control group, the positive rate of PCNA in SGC-7901 cells at 48h and 72h was significantly decreased (P <0.05 and P <0.01), and the cells in G0 + G1 phase were significantly increased (P <0.01 and P <0.05) There was no significant difference in the ratio of PCNA antibody positive cells between 24h and 36h. Compared with the control group, the positive rate of 12hPCNA antibody in HFE-145 cells in revolving group was significantly decreased (P <0.05), and the cells in G0 + G1 phase were significantly increased (P <0.05). CONCLUSIONS: Rotavator simulated cell cycle arrest and proliferation inhibition in SGC-7901 cells at 48h and 72h after simulated weightlessness. The gastric mucosal epithelial HFE-145 cells at 12h when the above changes.
