[目的]探讨钙敏感受体(Calcium-sensing receptor,CaR)在淤胆型肝炎小鼠肝组织中的表达。[方法]采用ANIT(80mg/kg,po)制备小鼠淤胆型肝炎模型,检测给予ANIT 24h、48h后小鼠血清ALT、ALP及BA水平,HE染色观察肝脏病理组织学改变,免疫组化检测CaR在淤胆型肝炎小鼠肝组织中的表达。[结果]给予ANIT 24h、48h后,小鼠血清ALT、ALP及BA水平逐渐升高,48h后明显上升;病理结果显示48h后,肝细胞坏死明显;免疫组化结果显示淤胆型肝炎小鼠肝组织中CaR的表达明显增强。[结论]CaR在淤胆型肝炎小鼠肝脏组织中存在高表达,且可能与淤胆的发生有关。
[Objective] To investigate the expression of Calcium-sensing receptor (CaR) in liver of mice with cholestatic hepatitis. [Methods] The mouse model of cholestatic hepatitis was established by ANIT (80mg / kg, po). The serum levels of ALT, ALP and BA were detected after ANIT for 24h and 48h. The pathological changes of liver were observed by immunohistochemistry To detect the expression of CaR in the liver of cholestatic hepatitis mice. [Results] The levels of ALT, ALP and BA in serum of the mice increased gradually after 48 hours and reached the peak at 48 hours after ANIT treatment. The pathological results showed that the necrosis of hepatocytes was obvious after 48 hours. The results of immunohistochemistry showed that the mice with cholestatic hepatitis The expression of CaR in liver tissue was significantly increased. [Conclusion] CaR is highly expressed in the liver of mice with cholestatic hepatitis and may be related to the occurrence of cholestasis.
