目的:探究替米沙坦对东莨菪碱模型小鼠学习记忆及脑内胆碱能神经的影响。方法:将小鼠按体质量随机分为5组:正常对照组(Sal+Veh)、东莨菪碱模型组(Sco+Veh)、多奈哌齐组(Sco+Ari)、替米沙坦高剂量组(Sco+Tel 0.70mg.kg-1)及低剂量组(Sco+Tel 0.35mg.kg-1),灌胃给药,除正常对照组腹腔注射生理盐水外,其他各组注射东莨菪碱。采用Morris水迷宫和Y迷宫试验评价学习记忆能力,并测定脑内乙酰胆碱(ACh)、乙酰胆碱酯酶(AChE)水平。结果:替米沙坦(0.70,0.35mg.kg-1.d-1)能显著缩短东莨菪碱模型鼠在隐藏平台试验的潜伏期,增加空间探索试验目标象限的停留时间百分率及穿台次数,增加Y迷宫试验的正确反应次数。替米沙坦还能显著抑制东莨菪碱模型鼠脑海马及皮层区AChE活性,增加ACh含量。结论:替米沙坦可通过抑制东莨菪碱模型小鼠脑内AChE活性增加ACh含量,改善学习记忆能力。
Objective: To investigate the effect of telmisartan on learning and memory and the cholinergic nerve in scopolamine model mice. Methods: The mice were randomly divided into five groups according to their body weight: Sal + Veh, Scopolamine (Sco + Veh), Donepezil (Sco + Ari) Tel 0.70mg.kg-1) and low-dose group (Sco + Tel 0.35mg.kg-1) were administered intragastrically. Except for the normal control group, normal saline was injected intraperitoneally, while other groups were injected with scopolamine. Morris water maze and Y-maze test were used to evaluate the learning and memory abilities, and the levels of acetylcholine (ACh) and acetylcholinesterase (AChE) in the brain were measured. RESULTS: Telmisartan (0.70,0.35mg.kg-1.d-1) significantly shortened the incubation period of scopolamine in the hidden platform test, increased the percentage of stay time and the number of piercing times in the target quadrant of the space exploration experiment, and increased Y Maze test the correct number of responses. Telmisartan can significantly inhibit scopolamine-induced AChE activity in the hippocampus and cortex and increase ACh content. Conclusion: Telmisartan can improve the ability of learning and memory by inhibiting AChE activity in brain of scopolamine model mice.
