克隆了一株抗全反式维甲酸(RA)分化诱导作用的人早幼粒白血病细胞HL-60的亚系HL-60/RA:其抗分化特性不同于肿瘤细胞对一般细胞毒抗癌药的抗药性。可不依赖RA而传代培养。抗性稳定至少18月之久。HL-60/RA的抗分化具有定向特异性:对粒系分化诱导剂具有交叉抗性。而对单核-巨噬细胞系分化诱导剂仍然敏感。说明HL-60粒系分化与单核-巨噬细胞系分化具有不同的调控机制。HL-60/RA的克隆为深入研究肿瘤细胞的抗分化作用以及研究分化诱导剂和细胞分化的调控机理提供了工具。
The sub-line HL-60 / RA, a human promyelocytic leukemia cell line HL-60 that has been induced by differentiation-inducing all-trans retinoic acid (RA), was cloned. Its anti-differentiation property is different from that of tumor cells on general cytotoxic anticancer drugs Resistance. Can not rely on RA and subculture. Resistance is stable for at least 18 months. The anti-differentiation of HL-60 / RA is directional-specific: cross-resistant to the granulocyte differentiation inducer. While still sensitive to monocyte-macrophage cell lineage-inducing agents. These results suggest that HL-60 has different regulatory mechanisms between myelomonocytic differentiation and monocyte-macrophage cell differentiation. The cloning of HL-60 / RA provides a tool to further study the anti-differentiation of tumor cells and to study the regulation mechanism of differentiation inducer and cell differentiation.
