目的:探讨成纤维细胞生长因子21(Fibroblast growth factor 21,FGF21)基因是否在心脏中表达及其表达水平,为进一步研究FGF21与动脉粥样硬化发生发展的关系提供理论基础。方法:对培养的乳鼠心肌细胞、大鼠心脏微血管内皮细胞进行RealTime-PCR定量检测,并与FGF21的主要来源肝脏进行比较,测定FGF21基因在心肌细胞与内皮细胞中的表达水平。结果:RealTime-PCR结果显示FGF21基因在心肌细胞与内皮细胞中均有表达,且ΔCt值肝脏显著低于心肌细胞与内皮细胞[(5.50592±0.18630)vs(11.26081±0.25024)vs(15.99978±0.17076),P<0.05]。结论:ΔCt值与样品中FGF21基因的表达成反比,即肝脏组织表达FGF21基因的水平高于心肌细胞与内皮细胞。此外本研究发现,心肌细胞与内皮细胞中均有FGF21基因表达,可能对心肌损伤起一定作用。
OBJECTIVE: To investigate whether FGF21 gene is expressed in the heart and its expression level, so as to provide a theoretical basis for further study on the relationship between FGF21 and the development of atherosclerosis. Methods: RealTime-PCR was used to detect the cardiomyocytes and rat cardiac microvascular endothelial cells. The expression of FGF21 gene in cardiomyocytes and endothelial cells was determined by real-time PCR and compared with the main source liver of FGF21. Results: Real-time PCR showed that FGF21 gene was expressed in both cardiomyocytes and endothelial cells, and the ΔCt value was significantly lower in liver than in cardiomyocytes and endothelial cells [(5.50592 ± 0.18630) vs (11.26081 ± 0.25024 vs (15.99978 ± 0.17076) , P <0.05]. CONCLUSION: The ΔCt value is inversely proportional to the expression of FGF21 gene in the sample, indicating that the level of FGF21 gene in liver tissue is higher than that of cardiomyocytes and endothelial cells. In addition, this study found that both cardiomyocytes and endothelial cells have FGF21 gene expression, may play a role in myocardial injury.
