目的观察恶性疟原虫海南分离株(FCC1/HN)裂殖子表面蛋白2(MSP2)融合HBsAg基因片断真核表达质粒pVXORFl-PfMSP2-HBS在小鼠骨骼肌的表达。方法KM小鼠分3组,用0.5%盐酸布比卡因对3组小鼠行预处理后,在同一部位Ⅰ组注射浓度为1μg/μl的重组质粒pVXORFl-PfMSP2-HBS100μl,Ⅱ组注射浓度为1μg/lμ的质粒pVXORFl-HBS100μl,Ⅲ组为正常对照组,注射pH7.4PBS100μl。分别在免疫后4周和8周用免疫组织化学实验检测小鼠注射部位肌肉组织中重组蛋白MSP2的表达。结果小鼠经DNA疫苗注免疫4周和8周后肌肉组织呈特异性阳性反应。结论重组质粒pVXORFl-PfMSP2-HBS免疫小鼠后,肌肉组织有重组蛋白MSP2的表达。
Objective To observe the expression of plasmid pVXORF1-PfMSP2-HBS in the skeletal muscle of mice induced by fusogenic surface protein 2 (MSP2) fused with HBsAg gene of Plasmodium falciparum Hainan (FCC1 / HN). Methods KM mice were divided into three groups. After pretreatment with 0.5% bupivacaine hydrochloride in three groups, 100 μg recombinant plasmid pVXORF1-PfMSP2-HBS (1 μg / μl) was injected in group Ⅰ at the same site. 100 μl of plasmid pVXORF1-HBS (1 μg / μl), group Ⅲ (normal control group) and 100 μl of pH 7.4 PBS. Immunohistochemistry was used to detect the expression of recombinant protein MSP2 in the muscle of mice at 4 and 8 weeks after immunization. Results The mice were immunized 4 weeks and 8 weeks after DNA vaccination with muscle tissue showed a specific positive reaction. Conclusion The recombinant plasmid pVXORF1-PfMSP2-HBS immunized mice, the muscle tissue recombinant protein MSP2 expression.
