目的对产于我国甘肃省的中药秦艽的3种基源植物秦艽、麻花秦艽与小秦艽进行RAPD分析,建立具有鉴别意义的DNA指纹图谱。方法采用RAPD分子标记对秦艽的基因组DNA进行PCR扩增,利用NTSYSpc 2.1和Popgene 3.2软件分析3种秦艽间的亲缘关系,构建树系图。结果从80条RAPD引物中筛选出4条具有鉴别意义的多态性引物,在3种秦艽中共得到39个DNA条带,其中共有条带2条,多态条带37条。据此获得了能有效区别3种秦艽的多态性RAPD指纹谱。结论甘肃产秦艽的3种基源植物遗传分化明显,RAPD方法能有效地将它们区分鉴别。
Objective To carry out RAPD analysis on the three basic plant species of Gentiana macrophylla, Gentiana straminea and Gentiana macrophylla, which are produced in Gansu Province of China, and to establish a DNA fingerprinting with distinct significance. Methods RAPD molecular markers were used to amplify the genomic DNA of Gentiana macrophylla and NTSYSpc 2.1 and Popgene 3.2 software were used to analyze the genetic relationships among the three species of Gentiana and construct a tree line map. Results Four polymorphic primers were screened out from 80 RAPD primers. Thirty-nine DNA bands were obtained in the three species of Gentiana. Among them, there were 2 total bands and 37 polymorphic bands. Based on this, we obtained the polymorphic RAPD fingerprinting that can effectively distinguish the three species of Gentiana. Conclusion The genetic differentiation of the three basic plant species in G. gentiana produced in Gansu Province is obvious. The RAPD method can distinguish them effectively.
