目的:探讨顺铂(DDP)作用于人宫颈癌SiHa细胞时肺癌肿瘤抑制因子1(TSLC1)基因的表达情况。方法:噻唑蓝(MTT)法测定不同浓度(5、10、15、20、25μg/ml)DDP干预SiHa细胞后的抑制率;采用实时荧光定量(RT-PCR)技术检测不同浓度的DDP处理前、后TSLC1基因mRNA表达水平的变化。结果:DDP呈剂量-时间依赖方式抑制SiHa细胞的生长(P<0.01);且随着DDP浓度的增高,TSLC1 mRNA表达上调(P<0.05)。结论:DDP抑制人宫颈癌SiHa细胞生长的机制可能与上调TSLC1基因mRNA的表达有关。
Objective: To investigate the expression of tumor suppressor 1 (TSLC1) gene in human cervical cancer SiHa cells induced by cisplatin (DDP). Methods: The inhibitory rates of SiHa cells treated with DDP at different concentrations (5, 10, 15, 20, 25μg / ml) were determined by MTT assay. Before treatment with different concentrations of DDP, , After TSLC1 gene mRNA expression level changes. Results: DDP inhibited the growth of SiHa cells in a time-and dose-dependent manner (P <0.01). The expression of TSLC1 mRNA was up-regulated with the increase of DDP concentration (P <0.05). Conclusion: The mechanism of DDP inhibiting the growth of human cervical cancer SiHa cells may be related to up-regulating TSLC1 gene mRNA expression.
