目的:探讨白细胞介素10(IL-10)对慢性心力衰竭(CHF)患者外周血单个核细胞(PBMCs)促炎性细胞因子分泌的影响。方法:15例CHF患者和15例健康人清晨采外周静脉血8ml,肝素抗凝,用淋巴细胞分离液分离出PBMCs。分3组对PBMCs进行培养:用1640培养基培养作为空白组;用含脂多糖(LPS,2mg/L)的1640培养基作为LPS组;用含LPS(2mg/L)和IL-10(20μg/L)的1640培养基培养作为IL-10组。把含有PB-MCs的培养板放在含5%二氧化碳的孵箱中37℃孵育24h,收集培养液离心,吸取上清液。检测肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素6(IL-6)水平。结果:CHF患者无论是空白组、LPS组还是IL-10组,TNF-α、IL-1β和IL-6水平均比健康人增高,均差异有统计学意义(均P<0.01)。LPS组比空白组TNF-α、IL-1β、IL-6升高,IL-10组比LPS组TNF-α、IL-1β、IL-6降低,均差异有统计学意义(P<0.05,P<0.01)。结论:CHF患者PBMCs分泌炎性细胞因子增加,而IL-10具有明显的抑制作用。
Objective: To investigate the effect of interleukin-10 (IL-10) on the secretion of proinflammatory cytokines by peripheral blood mononuclear cells (PBMCs) from patients with chronic heart failure (CHF). Methods: Peripheral blood samples from 15 CHF patients and 15 healthy individuals were collected in the early morning for 8ml. Heparin was anticoagulated and PBMCs were isolated by lymphocyte separation. PBMCs were cultured in three groups: 1640 medium was used as a blank group; 1640 medium containing lipopolysaccharide (LPS, 2 mg / L) was used as LPS group; LPS (2 mg / L) and IL- / L) 1640 medium as an IL-10 group. Plates containing PB-MCs were incubated in an incubator containing 5% carbon dioxide at 37 ° C for 24 hours. The culture broth was centrifuged and the supernatant was aspirated. The levels of tumor necrosis factor alpha (TNF-α), interleukin 1β (IL-1β) and interleukin 6 (IL-6) were detected. Results: The levels of TNF-α, IL-1β and IL-6 in CHF patients were significantly higher than those in healthy subjects (both P <0.01), no matter in blank control group, LPS group or IL-10 group. The levels of TNF-α, IL-1β and IL-6 in LPS group were significantly higher than those in control group, while the levels of TNF-α, IL-1β and IL-6 in IL-10 group were lower than those in LPS group P <0.01). Conclusion: The inflammatory cytokines secreted by PBMCs in CHF patients increased, while IL-10 had a significant inhibitory effect.
