目的探讨在ICSI周期中,不同的培养成分对人MⅠ期未成熟卵母细胞的应用价值。方法随机将ICSI周期中316枚MⅠ期未成熟卵母细胞分成四组:A组(HTF培养液),B组(A组+卵泡液),C组(A组+颗粒细胞),D组(A组+卵泡液+颗粒细胞)。分别记录各组未成熟卵母细胞的成熟率、受精率、优质胚胎率及可用胚胎率,并进行组间对比,所得数据采用SPSS13.0软件进行统计学处理。计数资料比较采用χ2检验,计量资料采用t检验,P<0.05有统计学意义。结果 1C组中24h成熟率、优质胚胎率和可用胚胎率均高于其它各组数值,比较均有统计学意义(P<0.05);2B组中24h成熟率、优质胚胎率和可用胚胎率均高于A组中各数值,比较均有统计学意义(P<0.05);3B组中24h成熟率、优质胚胎率和可用胚胎率与C组中各值比较无统计学意义(P>0.05);结论培养基中添加卵泡液及颗粒细胞对MⅠ期未成熟卵母细胞的成熟及发育潜能有重要作用。
Objective To explore the value of different culture components in human M Ⅰ immature oocytes during ICSI cycles. Methods 316 MⅠ immature oocytes in the ICSI cycle were randomly divided into four groups: group A (HTF), group B (group A + follicular fluid), group C (group A + granulocyte), group D A group + follicular fluid + granulosa cells). The maturation rate, fertilization rate, high quality embryo rate and available embryo rate of immature oocytes in each group were recorded and compared between groups. The data were analyzed by SPSS 13.0 software. Counting data were compared using χ2 test, measurement data using t test, P <0.05 was statistically significant. Results The rate of 24 h ripening, high quality embryo and available embryo in 1C group were higher than those in other groups (P <0.05). In group 2B, the rate of 24 h ripening, high quality embryo and available embryo (P <0.05). There was no significant difference in 24h maturation rate, high quality embryo rate and available embryo rate in group 3B compared with those in group C (P> 0.05) ; Conclusion The addition of follicular fluid and granulosa cells in culture medium plays an important role in the maturation and developmental potential of M Ⅰ immature oocytes.
