本文报道了用[~3H-甲基]-甲硫氨酸代替S-腺苷-L甲硫氨酸作为甲基供体,在家蚕细胞质多角体病毒(简称CPV)颗粒所带的RNA多聚酶催化下于体外合成~3H-mRNA。体外合成的~3H-CPV mRNA经 DEAE-Sephadex A-25柱层析分离纯化。用核糖核酸酶P_1和蛇毒磷酸二酯酶降解、纸电泳分离,结果表明甲硫氨酸的[~3H-甲基]已参入CPV-RNA的5-末端。即使在S-腺苷-L-甲硫氨酸的存在下,[~3H-甲基]-甲硫氨酸对于CPV-mRNA的5’-末端罩式结构的形成仍具有甲基供体的功能。
In this paper, the use of [~ 3H-methyl] -methionine instead of S-adenosyl-L-methionine as a methyl donor has been reported to catalyze the transcription of RNA polymerase by RNA polymerase in silkworm cytoplasmic polyhedrosis virus (CPV) ~ 3H-mRNA was synthesized in vitro. The ~ 3H-CPV mRNA synthesized in vitro was purified by DEAE-Sephadex A-25 column chromatography. It was degraded by RNase P_1 and PHD, and electrophoresed on paper. The results showed that [~ 3H-methyl] of methionine had been involved in the 5-terminal of CPV-RNA. The formation of the 5 ’-end capping structure of CPV-mRNA by [~ 3H-methyl] -methionine, even in the presence of S-adenosyl-L-methionine, Features.
