目的:探讨腺相关病毒介导的CD151在小型猪急性心肌梗死(AMI)后对心肌细胞凋亡和Bax/Bcl-2蛋白的影响。方法:建立猪AMI模型,随机分为4组,分别给心肌内注入腺相关病毒介导的GFP基因(rAAV-GFP组)、腺相关病毒介导的CD151基因(rAAV-CD151组)、腺相关病毒介导的反义CD151基因(rAAV-an-tiCD151组)、假手术组(Control组)。AMI后2个月处死,TUNEL法检测心肌细胞凋亡,WesternBlot法检测各组CD151、Bcl-2、Bax蛋白的表达。结果:凋亡指数,rAAV-CD151组(3.3±0.4)小于rAAV-GFP组(9.2±1.3)及rAAV-antiCD151组(10.1±1.5)(P<0.05);rAAV-CD151组略多于Control组(2.4±0.8),2组差异无统计学意义,rAAV-CD151组CD151表达高于其他组,Bax/Bcl-2(1.13±0.11)较rAAV-GFP组(3.21±0.12)及rAAV-antiCD151组(3.53±0.09)明显降低(P<0.05),与Control组(0.93±0.04)比较差异无统计学意义(P>0.05)。结论:腺相关病毒介导的CD151基因治疗能明显减少心肌细胞凋亡,改善心功能,这可能是由于CD151减低Bax/Bcl-2比值的结果。
AIM: To investigate the effect of adeno-associated virus-mediated CD151 on cardiomyocyte apoptosis and Bax / Bcl-2 protein after mini-swine acute myocardial infarction (AMI). Methods: Pig AMI models were established and randomly divided into 4 groups. Adeno-associated virus-mediated GFP gene (rAAV-GFP group), adeno-associated virus-mediated CD151 gene (rAAV-CD151 group) Virus-mediated antisense CD151 gene (rAAV-an-tiCD151 group), sham operation group (Control group). Two weeks after AMI, cardiomyocyte apoptosis was detected by TUNEL method. The expressions of CD151, Bcl-2 and Bax in each group were detected by Western blot. Results: The apoptotic index in rAAV-CD151 group was significantly lower than that in rAAV-GFP group (9.2 ± 1.3) and rAAV-antiCD151 group (10.1 ± 1.5) (P <0.05) (2.4 ± 0.8). There was no significant difference between the two groups. The expression of CD151 in rAAV-CD151 group was higher than that in other groups. The level of Bax / Bcl-2 was significantly higher in rAAV-antiCD151 group than in rAAV-GFP group (1.13 ± 0.11 vs 3.21 ± 0.12) (3.53 ± 0.09) (P <0.05), but there was no significant difference between the control group and the control group (0.93 ± 0.04) (P> 0.05). Conclusions: Adeno-associated virus-mediated CD151 gene therapy can significantly reduce cardiomyocyte apoptosis and improve cardiac function, which may be due to the decrease of Bax / Bcl-2 ratio by CD151.
