目的 观察卵巢癌抗独特型单链抗体 6B11ScFv与鼠粒细胞巨噬细胞集落刺激因于(murine granulocvte-macrophage colony-stimulating factor.mGM-CSF)的融合蛋白(6B11mGM)在小鼠体内诱导抗肿瘤免疫反应情况,探讨此种融合蛋白作为卵巢癌疫苗的可能性及适当的免疫方法。方法 用融合蛋白6B11mGM免疫BALB/c小鼠,制备抗血清。采用间接ELISA及免疫流式方法分析抗血清特征。结果 经ELISA检测,融合蛋白6B11mGM免疫小鼠后,可诱导机体产生AB3。同时可刺激小鼠脾脏淋巴细胞CD4+细胞明显升高,CD8+细胞略有升高。结论 融合蛋白6B11mGM可诱导机体产生特异体液免疫和细胞免疫反应,为抗独特型卵巢癌疫苗用于临床奠定了基础。
Objective To observe the anti-tumor immunity induced by a murine granulocvte-macrophage colony-stimulating factor (mGM-CSF) fusion protein (6B11mGM) Response to explore the possibility of this fusion protein as an ovarian cancer vaccine and appropriate immunization methods. Methods BALB / c mice were immunized with fusion protein 6B11mGM to prepare antiserum. Antisera characteristics were analyzed by indirect ELISA and immunofluorescence methods. Results After ELISA, the mice immunized with the fusion protein 6B11mGM could induce the body to produce AB3. At the same time can stimulate the mouse spleen lymphocytes CD4 + cells was significantly increased, CD8 + cells slightly increased. Conclusion The fusion protein 6B11mGM can induce specific humoral and cellular immune responses and lay the foundation for the anti-idiotypic ovarian cancer vaccine.
