目的研究分析人乳头瘤病毒(HPV)16、18感染与宫颈病变组织γ干扰素(IFN-γ)及白细胞介素-10(IL-10)表达的关系,为临床诊疗提供参考。方法选择2015年1月-2015年7月在佛山市妇幼保健院妇科进行诊治的126例HPV感染者作为研究对象。根据患者HPV的DNA检测分型结果,将其分成HPV16感染组42例,HPV18感染组42例,以及非高危型HPV感染(NHRT-HPV)组42例,比较各组宫颈病变组织的IFN-γ、IL-10表达,Toll样受体4(TLR4)信号通路相关指标,以及线粒体腺苷酸转运体(ANT)、程序性死亡分子配体2(PD-L2)及凋亡抑制基因(Survivin)的表达,并分析HPV感染与上述指标的相关性。结果各组IFN-γ及IL-10的mRNA及蛋白水平比较,差异存在统计学意义(P<0.05)。其中HPV16组和HPV18组IFN-γ的mRNA及蛋白水平明显低于NHRT-HPV组,IL-10则明显高于NHRT-HPV组(P<0.05)。各组TLR4、依赖髓样分化因子(My D88)及核因子kB(NF-k B)的mRNA及蛋白水平比较,差异存在统计学意义(P<0.05)。HPV16组和HPV18组TLR4、My D88及NF-k B的mRNA及蛋白水平均明显高于NHRT-HPV组(P<0.05)。各组ANT、PD-L2及Survivin的mRNA及蛋白水平比较,差异存在统计学意义(P<0.05)。HPV16组和HPV18组ANT的mRNA及蛋白水平明显低于NHRTHPV组,PD-L2及Survivin的mRNA及蛋白水平明显高于NHRT-HPV组(P<0.05)。HPV感染与IL-10、TLR4、My D88、NFk B、PD-L2及Survivin呈正相关,与IFN-γ、ANT呈负相关。结论 HPV感染与免疫功能指标,TLR4信号通路相关指标以及细胞增殖相关基因之间存在紧密联系,其中HPV16、18感染与IL-10、TLR4、My D88、NF-k B、PD-L2及Survivin呈正相关,与IFN-γ、ANT呈负相关。
Objective To study the relationship between the expression of human interferon-gamma (IFN-γ) and interleukin-10 (IL-10) in cervical lesions and human papillomavirus (HPV) 16,18 infection and to provide references for clinical diagnosis and treatment. Methods From January 2015 to July 2015, 126 cases of HPV infection in gynecology of Foshan MCH hospital were selected as the research object. According to the DNA typing results of patients with HPV, 42 cases of HPV16 infection, 42 cases of HPV18 infection and 42 cases of non-high risk HPV infection (NHRT-HPV) were divided into three groups: IL-10, Toll-like receptor 4 (TLR4) signaling pathway, as well as mitochondrial adenylate transporter (ANT), PD-L2 and Survivin, The correlation between HPV infection and the above indexes was analyzed. Results The mRNA and protein levels of IFN-γ and IL-10 in each group were significantly different (P <0.05). The mRNA and protein levels of IFN-γ in HPV16 and HPV18 groups were significantly lower than those in NHRT-HPV group and IL-10 group (P <0.05). The mRNA and protein levels of TLR4, My D88 and NF-κB in each group were significantly different (P <0.05). The mRNA and protein levels of TLR4, My D88 and NF-κB in HPV16 group and HPV18 group were significantly higher than those in NHRT-HPV group (P <0.05). The mRNA and protein levels of ANT, PD-L2 and Survivin in each group were significantly different (P <0.05). The mRNA and protein levels of ANT in HPV16 and HPV18 groups were significantly lower than those in NHRTHPV group. The mRNA and protein levels of PD-L2 and Survivin were significantly higher than those in NHRT-HPV group (P <0.05). HPV infection was positively correlated with IL-10, TLR4, My D88, NFk B, PD-L2 and Survivin, but negatively correlated with IFN-γ and ANT. Conclusion HPV infection is closely related to immune function, TLR4 signaling pathway and cell proliferation related genes. HPV16,18 infection is positively correlated with IL-10, TLR4, My D88, NF-κB, PD-L2 and Survivin Related, and IFN-γ, ANT was negatively correlated.
