目的 :研究血清样品稀释度、温度、离子强度以及样品检测间隔时间对血清抗心磷脂抗体 (ACA)检测结果的影响。方法 :采用酶联免疫吸附测定法 (ELISA)检测ACA。结果 :血清样品最佳稀释度为 1∶5 0 ;37℃时光吸收值比 2 2℃和 2 5℃的光吸收值显著下降 ;15mol LNaCl可明显抑制ACA与抗原结合 (P <0 0 1) ,抑制率平均为 45 4% ;包被 14d以上的检测结果明显高于 1d和 7d的测定结果 (P <0 0 5 )。结论 :血清ACA检测结果受血清样品稀释度、温度、离子强度及包被时间影响。
Objective: To study the influence of serum sample dilution, temperature, ionic strength and sample detection interval on the results of serum anticardiolipin antibody (ACA). Methods: ACA was detected by enzyme-linked immunosorbent assay (ELISA). Results: The optimal dilution of serum sample was 1: 50; the absorbance of light at 37 ℃ was significantly lower than that at 22 ℃ and 25 ℃; 15mol LNaCl significantly inhibited the binding of ACA to antigen (P <0.01) , And the average inhibition rate was 45 4%. The test results of more than 14 days after coating were significantly higher than those of 1 day and 7 days (P <0 05). Conclusion: The result of serum ACA is affected by the dilution, temperature, ionic strength and coating time of serum samples.
